Saunders failed a voluntary drugs test after testing positive for the banned stimulant oxilofrine, and the boxing commission in Massachusetts voted to deny him a license for the bout.
Saunders reacted by saying: “You can s**k my p***k.”
Saunders became the WBO middleweight world champion when he beat Andy Lee in December 2015.
He previously held the European, British, and Commonwealth middleweight titles.
This episode is the latest in a string of controversies for Saunders, who was fined £100,000 (N47.7m) by the British Boxing Board of Control last month after a video surfaced of him asking a woman to perform a sex act on a passer-by in exchange for drugs.
Unbeaten in 26 fights, he was due to defend his title against American Andrade in Boston on 20 October, but is now set to be stripped of his world title.
